Properties and action of Acetyl glucosamine in cosmetics
Acetyl glucosamine (NAG), an amino monosaccharide (simple sugar), is the basic monomeric unit of chitin. One of the most abundant natural polysaccharides, chitin is found in the cell walls of fungi, insect exoskeletons, and crustacean shells. In humans, NAG can be identified as part of the glycosaminoglycans of the intercellular matrix, hyaluronic acid. As a major component of connective tissue, hyaluronic acid is abundant in the dermal layer of the skin, and NAG is its precursor, i.e., it increases its production.
Thus, NAG is considered a biologically identical ingredient that supports the barrier properties of the skin.
The natural role of acetyl glucosamine in the skin:
- Inhibits the production and uneven distribution of melanin in melanocyte culture
- has the ability to inhibit tyrosinase glycosylation, which limits the degree of pigmentation
- affects the signs of photoaging caused by UV radiation
- as a natural structural component of hyaluronic acid, increases its production by skin cells, and also participates in the healing process
- antioxidant.
Given the fact that pure acetyl glucosamine is a sensitive and poorly stable ingredient, its synthetic or biofermented derivative, known as N-acetyl-D-glucosamine, has proven to be more highly effective in the cosmetic realm. We offer a vegan stable form of glucosamine derived from the biofermentation process, Hyafactor-NAG.
Cosmetic properties of Hyafactor-NAG
- reduces hyperpigmentation, brightens and evens overall skin tone
- anti-ageing component, which helps to smooth existing and emerging wrinkles
- has a pronounced anti-inflammatory effect
- is used in the complex treatment of common acne of mild to moderate degree
- provides optimal hydration, contributing to skin moisturization and elasticity
- "non-acidic ingredient with a mild exfoliating effect to regulate keratinization
- fights photoaging
- skin firming
- reduces sensory irritation and associated discomfort
- External supply of NAG supports collagen production.
Efficacy study
Skin brightening effect
Below you can see graphs showing the effect of Hyafactor-NAG on melanin formation and tyrosinase activity.

Thus, the efficacy of NAG to reduce hyperpigmentation when applied topically has been demonstrated.
Exfoliating effect on the skin
NAG is a natural regulator of exfoliation during keratinocyte metabolism. A glycoprotein present on the surface of keratinocytes causes these keratinized keratinocytes to bind together and are difficult to peel off when metabolism is impaired. NAG, on the other hand, can improve and maintain glycoprotein metabolism on the surface of keratinocytes in such a way that the natural exfoliation of cutin makes the skin smoother and softer.

Benefits of using NAGs
The importance of including NAG in cosmetics is based not only on its safety and chemical neutrality, but also on its proven ability to reduce inflammation and pigmentation. If the skin, due to acquired sensitivity, reacts to acids (AHA and BHA), NAG can be an alternative compound to achieve skin rejuvenating effects. Due to the high affinity, cosmetics with NAGs are well tolerated by the skin.
Dose-dependent moisturizing effect

Synergistic effects:
- The combination of 2% NAG with 4% niacinamide is used to reduce hyperpigmentation and even out skin tone
- The combination of NAG with retinoids or carotenoids (found in sea buckthorn, wheat germ oil, etc.) stimulates the production of native hyaluronic acid by the skin.
The high level of tolerance, in addition to relative ease of preparation and stability, makes NAG an ideal cosmetic ingredient for use in skin care products and anti-aging therapies.
